DUONEX-30/40 (DULOXETINE TABLETS)
DESCRIPTION
Duloxetine is a selective serotonin and norepinephrine reuptake inhibitor (SSNRI) for oral administration. Its chemical designation is (+)-(S)-N-methyl-γ-(1-naphthyloxy)-2-thiophenepropylamine hydrochloride. The empirical formula is C18H19NOS•HCl.
DOSAGE
Initial dose: 20 mg orally twice a day. Maintenance dose: 60 mg per day, given either once a day or as 30 mg orally twice a day. Maximum dose: 120 mg orally per day
CLINICAL PHARMACOLOGY
Duloxetine inhibits the reuptake of serotonin and norepinephrine (NE) in the central nervous system. Duloxetine increases dopamine (DA) specifically in the prefrontal cortex where there are few DA reuptake pumps via the inhibition of NE reuptake pumps (NET) thus allowing greater diffusion of DA in this brain region
INDICATIONS
Major Depressive Disorder ,Generalized Anxiety Disorder ,Diabetic Peripheral Neuropathy,Fibromyalgia ,Chronic Musculoskeletal Pain.
CONTRA INDICATIONS
The use of MAOIs intended to treat psychiatric disorders with CYMBALTA or within 5 days of stopping treatment with CYMBALTA is contraindicated because of an increased risk of serotonin syndrome. The use of CYMBALTA within 14 days of stopping an MAOI intended to treat psychiatric disorders is also contraindicated.
